Identifying spatial immuno-oncological parameters of mouse tumors using Imaging Mass Cytometry

Abstract Identification of immuno-oncological (IO) processes that dictate tumor growth, metastasis, and immune response is essential for selecting promising drug candidates in preclinical models. Imaging Mass Cytometry™ (IMC™) a proven high-plex imaging technology enables deep characterization the complexity tissue without disrupting spatial context while eliminating artifacts caused by autofluorescence. Here, we showcase Maxpar® OnDemand™ Mouse Immuno-Oncology IMC Panel Kit application on mouse tissues. We compiled 33-plex antibody panel to quantitatively assess IO-related applied it microarray containing large variety tumors. digitized data from tissues using Hyperion™ System generated images demonstrating detailed layout microenvironment (TME). further conducted single-cell analysis identify specific populations cells TME. The IO successfully identified pathophysiological such as cell infiltration activation, signaling pathway biomarkers epithelial-to-mesenchymal transition (EMT), metabolic activity, TME architecture. Single-cell several highly relevant types separated distinct cellular clusters representing tumor, immune, stromal, vascular cells. Our quantitative composition revealed critical insights regarding prognostic parameters metastatic growth potential activity infiltrates. For Research Use Only. Not use diagnostic procedures.
